Rate of a reaction depends upon the concentration of the reactant as the concentration of the reactant decreases , the rate also decreases. As a result, for the reactant, the rate is indicated by a negative sign .

the main reason behind is that the rate of rxn keep on fluctuating with respect to time due to the concentration of reactant and product hence rate of rxn not remain constant throughout the rxn.
