Hershey and chase = they infected E.coli with bacteriophage and used radioactive sulphur ,radioactive phosphorus and the step were infection, blending, centrifugation and As the DNA contain phosphorus and protein contain sulphur. So at last after all steps they got DNA contained phosphorus, which shows that the DNA is transferred not protein. 

Hence it was unequivocal proof that DNA is the genetic material.
